Fila covers both a physical queue of people (fare la fila = to queue up) and a row of seats in a cinema or theatre. Fare la fila is the standard expression where English speakers say 'stand in line.' The plural file is regular. Note the phrase in fila indiana (single file, literally 'Indian file').
